Diversity and Evolution of the Phenazine Biosynthesis Pathway
2010
ABSTRACT Phenazines are versatile secondary metabolites of bacterial origin that function in biological control of plant pathogens and contribute to the ecological fitness and pathogenicity of the producing strains. In this study, we employed a collection of 94 strains having various geographic, environmental, and clinical origins to study the distribution and evolution of phenazine genes in members of the genera Pseudomonas , Burkholderia , Pectobacterium , Brevibacterium , and Streptomyces . Our results confirmed the diversity of phenazine producers and revealed that most of them appear to be soil-dwelling and/or plant-associated species. Genome analyses and comparisons of phylogenies inf…
Diastereo- and enantioselective synthesis of orthogonally protected 2,4-diaminocyclopentanecarboxylates: a flip from beta-amino- to beta,gamma-diamin…
2007
Conformationally restricted, orthogonally protected 2,4-diaminocarboxylates with a cyclopentane skeleton were efficiently synthesized from beta-lactam 6, the syntheses involving strategies of diastereoselective epoxidation of the beta-lactam and the corresponding monoprotected amino esters with opposite selectivities followed by regioselective opening of the oxirane ring with sodium azide. The enantiomers were also prepared. This new class of compounds can be regarded not only as conformationally constrained beta,gamma-diamino acid derivatives but also as potential functionalized carbocyclic nucleoside precursors.
Estimating the contribution of organic acids to northern hemispheric continental organic aerosol
2015
Using chemical ionization mass spectrometry to detect particle-phase acids (acid-CIMS) and aerosol mass spectrometry (AMS) measurements from Colorado, USA, and two studies in Hyytiala, Finland, we quantify the fraction of organic aerosol (OA) mass that is composed of molecules with acid functional groups (facid). Molecules containing one or more carboxylic acid functionality contributed approximately 29% (45-51%) of the OA mass in Colorado (Finland). Organic acid mass concentration correlates well with AMS m/z 44 (primarily CO2+), a commonly used marker for highly oxidized aerosol. Using the average empirical relationship between AMS m/z 44 and organic acids in these three studies, together…
Polyamine conjugates of stigmasterol.
2012
Abstract Three new polyamine conjugates with stigmasterol [(3β,22 E )-stigmasta-5,22-dien-3-ol] were synthesized and subjected to basic antimicrobial and cytotoxic tests. The conjugate derived from spermine, (3β,22 E )-stigmasta-5,22-dien-3-yl 4(12-amino-4,9-diaza-dodecylamino)-4-oxobutanoate ( 5c ), displayed considerable antimicrobial activity on Staphylococcus aureus at low concentration (50 μg mL −1 ). The cytotoxic activity was tested on cells of human T-lymfoblastic leukemia (IC 50 = 35.8 ± 10.3 μM ( 5c ) and IC 50 = 35.9 ± 5.7 μM ( 5b )) and normal human fibroblasts (IC 50 = 38.0 ± 2.8 μM ( 5c ) and IC 50 = 45.5 ± 1.9 μM ( 5b )). Conjugate 5a displayed no activity in both tests.
Validation of a novel automatic deposition of bacteria and yeasts on MALDI target for MALDI-TOF MS-based identification using MALDI Colonyst robot
2017
Matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS) -based identification of bacteria and fungi significantly changed the diagnostic process in clinical microbiology. We describe here a novel technique for bacterial and yeast deposition on MALDI target using an automated workflow resulting in an increase of the microbes' score of MALDI identification. We also provide a comparison of four different sample preparation methods. In the first step of the study, 100 Gram-negative bacteria, 100 Gram-positive bacteria, 20 anaerobic bacteria and 20 yeasts were spotted on the MALDI target using manual deposition, semi-extraction, wet deposition onto 70% formic …
A FRET-based probe for fluorescence sensing of sulfide/sulfite analytes, using a novel long-wavelength water-soluble 7-hydroxycoumarin as report…
2015
International audience; A FRET-based fluorescent probe for the detection of sulfide and/or sulfite in aqueous buffer, was constructed by connecting the quencher moiety nitrobenzofurazan (7-nitro-1,2,3-benzoxadiazole, NBD) to a water-soluble 3-(2-benzimidazolyl)-7-hydroxycoumarin carboxylic acid through a piperazine linker. This probe exhibits good selectivity and high sensitivity for sulfite and sulfide over cysteine and other potential analytes. Furthermore, this is one of the few examples of fluorogenic coumarins whose solubility in water is maintained upon its conversion into reaction-based fluorescent probes.
Combined Use of Structure Analysis, Studies of Molecular Association in Solution, and Molecular Modelling to Understand the Different Propensities of…
2021
The arrangement of hydroxyl groups in the benzene ring has a significant effect on the propensity of dihydroxybenzoic acids (diOHBAs) to form different solid phases when crystallized from solution. All six diOHBAs were categorized into distinctive groups according to the solid phases obtained when crystallized from selected solvents. A combined study using crystal structure and molecule electrostatic potential surface analysis, as well as an exploration of molecular association in solution using spectroscopic methods and molecular dynamics simulations were used to determine the possible mechanism of how the location of the phenolic hydroxyl groups affect the diversity of solid phases formed…
Stereocontrolled synthesis of D-α-hydroxy carboxylic acid from L-amino acids
1987
Abstract Optically active D-α-hydroxy carboxylic acids are obtained from L-amino acids via L-α-halocarboxylic acids and their stereoselective reaction with cesium p-nitrobenzoate.
ChemInform Abstract: Modified Photobehavior of Carboxylic Acid Derivatives Induced by Protonation.
1987
Abstract A series of carboxylic acid derivatives 1–6 containing a second interacting function have been converted into the corresponding O -protonated species or acylium ions upon treatment with 96% sulfuric acid or oleum, respectively, at room temperature. The resulting stable cations have been spectroscopically characterized and submitted to irradiation. Cis-trans photoisomerization was observed in the ionic speciesderived from trans -crotonic, fumaric and maleic acids, and maleic anhydride. By contrast protonated crotonolactone 5a+ was found to be photostable. Finally, protonated α-bromobutyrolactone 6a+ was clean and efficiently dehydrohalogenated to 5a+ upon irradiation. Since the norm…
Studies on the diurnal courses of the contents of abscisic acid, 1-aminocyclopropane carboxylic acid and its malonyl conjugate in needles of damaged …
1993
Summary The diurnal courses of the contents of 1-aminocyclopropane carboxylic acid (ACC), its malonyl conjugate (MACC) and abscisic acid (ABA) in the two-year-old needles of damaged and undamaged spruce trees (Picea abies L.) were investigated. The contents of ACC and MACC were significantly higher in the needles of the damaged trees as compared with the undamaged ones. In the needles of the damaged spruce trees, the ACC and MACC contents, and ABA contents as well, fluctuated to a greater extent than those in the undamaged spruce trees during the day. The relationships between ACC-ABA and MACC-ACC contents as well are also discussed.
